1. Trastuzumab triggered ADCC activity to MDA-MB-453 cells:

Figure 1. (Left) Effect of E/T (effector/target cells) ratio on ADCC. MDA-MB-453 cells (HER2 positive human breast cancer cell line) were incubated with the effector cells which are freshly purified human peripheral blood lymphocytes (PBL) at the indicated ratios with or without the presence of Trastuzumab for 4 hours at 37 °C/5%CO2. (Right) At the optimal E/T ratio - 25:1, the effector cells were incubated with MDA-MB-453 for 4 hours at 37 °C/5%CO2. Human Umbilical Vein Endothelial Cells (HUVEC, HER2 negative) and human whole IgG were used as the negative and isotype controls, respectively. The Trastuzumab-induced concentration-dependent cell lysis was detected using LDH kit. Absorbance was detected at 490 nm (formazan absorbance) and 650 nm (nonspecific absorbance).

2. Rituximab triggered CDC activity to Daudi cells
Figure 2. Daudi cells (CD20 positive B lymphoma cells) were pre-incubated with indicated concentration of Rituximab at RT for 15 min followed by addition of 10% NHS and incubated for another 45 min. The Rituximab-induced concentration-dependent cell lysis was analyzed with CellTiter-Glo? Luminescent Cell Viability Assay Kit.
