Description |
Protein kinase C ('PKC', EC 2.7.11.13) is a cyclic nucleotide-independent enzyme that phosphorylates serine and threonine residues in many target proteins. The PKC family has been divided into three groups differing in the enzymes' cofactor requirements: conventional (c)PKC isoforms (comprising α, βI (also known as β1), βII (also known as β2) and γ)), that require diacylglycerol (DAG), Ca2+, and phospholipid for activation; novel (n)PKC isoforms (comprising δ, ε, η (also known as PKC-L), θ and µ (the mouse homolog of human PKCµ, known as PKD)) that require DAG but not Ca2+; and atypical (a)PKC isoforms, namely ζ, ι and λ (the mouse homolog of human PKC ι) that require neither Ca2+ nor DAG. A new PKC member has recently been discovered and is referred to as PKCν. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role in cells. PKCζ (PKC zeta) exhibits a kinase activity which is independent of calcium and diacylglycerol but not of phosphatidylserine. Furthermore, it is insensitive to typical PKC inhibitors and cannot be activated by phorbol ester. Unlike the classical PKC isoenzymes, it has only a single zinc finger module. These structural and biochemical properties indicate that the zeta subspecies is related to, but distinct from other isoenzymes of PKC. GenScript Rabbit Anti-PKCζ Polyclonal Antibody is developed in rabbit using a synthetic peptide (KLH-coupled) derived from human PKCζ. |
Ig Subclass |
Rabbit IgG |
Storage |
The antibody is stable for 2-3 weeks if stored at 2-8°C. For long term storage, aliquot and store at -20°C or below. Avoid repeated freezing and thawing cycles. |
Specificity |
GenScript Rabbit Anti-PKCζ Polyclonal Antibody detects endogenous levels of total PKCζ protein. No cross-reactivity to other PKC isoforms is detected. |
Concentration |
0.5 mg/ml in PBS, pH 7.4, containing 30% glycerol and 0.02% sodium azide |
Immunogen |
Synthetic peptide (KLH-coupled) derived from human PKCζ |
Purification Method |
Immunoaffinity chromatography |
Applications |
Working concentrations for specific applications should be determined by the investigator. The appropriate concentrations may be affected by secondary antibody affinity, antigen concentration, the sensitivity of the method of detection, temperature, the length of the incubations, and other factors. The suitability of this antibody for applications other than those listed below has not been determined. The following concentration ranges are recommended starting points for this product. ELISA: 0.05-0.2 µg/ml Western blot: 0.5-2 µg/ml Other applications: user-optimized |
