PU.1 known as Spi1 and Sfpi1, is an ETS-domain transcription factor essential for the development of myeloid and B-lymphoid cells. In myeloid cells, PU.1 may regulate transcription of both FMS and CD18/11B proteins that are central to the osteoclast phenotype. The failure to express PU.1 might preclude development of osteoclasts, leading to arrested bone resorption and osteopetrosis. PU.1 expression increases with the induction of osteoclastogenesis by either 1,25-dihydroxyvitamin D3 or dexamethasone which confirms the association between PU.1 and osteoclast differentiation.
