The success of peptide drug often depends on modifying the naturally occurring peptide sequence chemically to improve their stability and bioavailability. The modification aiming for stability includes PEGylation and unnatural amino acids inclusion. The modification for bioavailability is achieved by their conjugation to surface receptor ligands (lectin, sugars, lipoproteins), antibodies, carrier peptides (TAT), or synthetic receptor ligands. In addition, addition of nuclear localization signals can improve subcellular targeting of the peptide drugs.
