Description |
Free fatty acid G protein coupled receptor family consists of four members and plays significant roles in nutrition regulation. GPR40 and GPR120 are activated by medium and long-chain FFAs, whereas GPR41 and GPR43 can be activated by short-chain FFAs. GPR40 is preferentially expressed in pancreatic beta-cells and mediates the majority of the effects of FFAs on insulin secretion. Researches show that GPR40 is a potential therapeutic target and plays an important role in the chain of events linking obesity and type2 diabetes. |

Figure 1. OA-induced concentration-dependent stimulation of intracellular calcium mobilization in CHO-K1/FFA1/Gα15 and CHO-K1/Gα15 cells. The cells were loaded with Calcium-4 prior to stimulation with a FFA1 receptor agonist, OA. The intracellular calcium change was measured by FlexStation. The relative fluorescent units (RFU) were plotted against the log of the cumulative doses (10-fold dilution) of OA (Mean ± SD, n = 2). The EC50 of OA on FFA1 co-expressing with Gα15 in CHO-K1 cells was 4.4 μM. The S/B of OA on FFA1 co-expressing with Gα15 in CHO-K1 cells was 15. |
