Problems
Full-length (or at least full-ORF) cDNAs are essential to the identification, cloning and characterization of functionally important genes. However, for many genes, isolating full-length cDNA is still a major technical challenge. Although cDNA libraries may be a highly efficient and convenient source of some cDNAs, the fragments derived from these libraries are usually very short and can only provide a little information. Covering a full-length cDNA requires several clones. This is especially important for rare species whose mRNA templates are hard to get and especially problematic for genes that have low abundance. Other factors, such as RNaseH activity and RNA secondary structure, can also affect the cloning of cDNA.
