Custom Monoclonal Antibody Services

We guarantee two positive clones for monoclonal antibodies developed in any host if peptide antigens were designed, synthesized, and conjugated by GenScript. For any protein antigen (2 mg, > 85% purity) we guarantee two positive clones with positive Western blot results for any host, or client pays only $800 set up fee.

1. SC1040: Partial Monoclonal Antibody Package

Phase I: Immunization and ELISA screening. Time: 8-10 weeks.

  • Conjugation: The peptide or small protein is conjugated to a carrier protein, such as KLH. Normally, 4 mg of protein immunogene or 5-10 mg of peptide is required.
  • Immunization: 5 Balb/c mice are immunized and boosted with standard immunization protocol.
  • Screening: Tail bleedings are screened by ELISA.

Phase II: Fusion and screening. Time: 4-8 weeks.

  • Fusion: Splenocyte fusions are performed on the mouse that responded the best to the immunization.
  • Expansion and screening: The hybridoma cells are expanded and screened by ELISA to obtain the highest-titer clones.

Phase III: Subcloning and delivery. Time: 4-6 weeks.

  • Subcloning: Subclone two positive parental clones (up to five) by limiting dilution and isotyped.
  • Delivery: One vial of cell culture for each clone and 5 ml of culture supernatants from the clones is delivered to the customer. Additional parental clones are available at an extra charge.

Total Time: 4-6 months.

2. SC1041: Complete Peptide Monoclonal Antibody Package

Phase I: Peptide synthesis. Time: 3-4 weeks.

  • 10 mg of 15 aa peptide with 90% purity is synthesized. 5 mg of free peptide will be delivered to the customer together with the antibody.

Phase II: Immunization and ELISA screening. Time: 8-10 weeks.

  • Conjugation: The peptide or small protein is conjugated to a carrier protein, such as KLH. Normally, 4 mg of protein immunogene or 5-10 mg of peptide is required.
  • Immunization: 5 Balb/c mice are immunized and boosted with standard immunization protocol.
  • Screening: Tail bleedings are screened by ELISA.

Phase III: Fusion and screening. Time: 4-8 weeks.

3. SC1301:  Phospho-Specific Monoclonal Antibody Package

Phase I: Peptide synthesis. Time: 3-4 weeks.

  • We will synthesize 10 mg of phospho-peptide and 5 mg non-phospho-peptide with up to 15 residues and 90% purity. 5 mg of free modified peptide and 3 mg of unmodified peptide will be delivered to the customer along with the final antibody product.

Phase II: Immunization and ELISA screening. Time: 8-10 weeks.

  • Conjugation: The modified peptide is conjugated to a carrier protein, such as KLH.
  • Immunization: 10 Balb/c mice are immunized and boosted with a standard immunization protocol.
  • Screening: Tail bleedings are screened by ELISA with phospho and non-phospho-peptide.

Phase III: Fusion and screening. Time: 4-8 weeks.

  • Fusion: Splenocyte fusions are performed on those mice that respond the best to the phospho-peptide in Phase II.
  • Expansion and screening: The hybridoma cells are screened by ELISA with phospho and non-phospho-petide and all positive clones are expanded into 24-well plates. The clones with best ELISA value and specificity to phospho-peptide are selected for subcloning.

Phase IV: Subcloning and delivery. Time: 4-6 weeks.

  • Subcloning: Hybridoma subclones are screened by ELISA with phospho and non-phospho-petide. We subclone up to five positive parental clones by limiting dilution, isotype the final positive supernatants, and expand and cryopreserve two subclones for each parental.
  • Delivery: Two vials of frozen cells or one flask of live cells and 5 ml of culture supernatants for a selected subclone. We will gladly provide subcloning for orders involving additional parental clones for an additional fee.

Total Time: 5-7 months.
