Human ether-a-gogo-related (hERG) ion channel has a major role during the repolarization of the cardiac action potential, and it is well established that blockade of this ion channel can lead to potentially lethal arrhythmias. Several drugs had to be withdrawn from the market due to their hERG cardiac toxicities. As a result, hERG (IKr) pre-clinical safety data is an essential part of any investigational new drug submission recommended in FDA ICH S7B. Recently, investigations suggest that non-hERG cardiac ion channels are also involved in cardiac toxicity.
