Description |
Human Bone Morphogenetic Protein-2 (BMP-2) is a bone-growth regulatory factor and a member of the transforming growth factor-β (TGF-β) superfamily of proteins. Human Bone Morphogenetic Protein-2 (BMP-2) is synthesized as large precursor molecules, which are cleaved by proteolytic enzymes. The active form can consist of a dimer of two identical proteins or as a heterodimer of two related bone morphogenetic proteins. Recombinant human Bone Morphogenetic Protein-2 (BMP-2) produced in E. coli is a homodimeric, non-glycosylated polypeptide chain containing 115 amino acids and having a molecular mass of 26,018 Da. Human Bone Morphogenetic Protein-2 (BMP-2) is purified by proprietary chromatographic techniques. |

rHuBMP-2 is fully biologically active when compared to standard. The ED50 as determined by the cytolysis of MC3T3-E1 cells is less than 50 ng/ml corresponding to a specific activity of 2.0 x 104 IU/mg. |
Storage |
Lyophilized recombinant human Bone Morphogenetic Protein-2 (BMP-2) remains stable at room temperature for three weeks, but it is best stored desiccated below -18°C. Upon reconstitution rHuBMP-2 should be stored at 4°C for up to seven days. For long term storage it is recommended that a carrier protein (0.1% HSA or BSA) be added. Avoid repeated freeze-thaw cycles. |
Formulation |
The protein was lyophilized after extensive dialysis against 50mM Acetic acid. |
Reconstitution |
It is recommended that the lyophilized rHuBMP-2 be reconstituted in sterile 20 mM HAc (acetic acid) not less than 100 μg/ml, which can then be further diluted to other aqueous solutions. |
Sequence analysis |
The sequence of the first five N-terminal amino acids has been found to be Met-Gln-Ala-Lys-His. |
