Description
High Range EasyWestern protein standard is a lyophilized mixture of five highly purified recombinant proteins ranging in size from 60 KD to 200 KD. Each of them contains an IgG binding domain. The high Range EasyWestern protein standard binds to the primary and/or the secondary antibody in the Western blot procedure. It enables direct visualization of both the standard and the sample on the same Western blot membrane without additional reagent.

Calculation
The apparent molecular weights of its five protein bands are 60 KD, 85 KD, 120 KD, 160 KD and 200 KD, respectively.
Note
1. Non-BSA based blocking reagents (such as 5% skimmed milk) are recommended for blocking.
2. For best results, the loading volume of the marker should be optimized for different experiment conditions (from 2.5 µl to 10 µl).
3. This marker binds to the constant region of IgG, so protein A or protein G conjugates cannot be used as the secondary antibody.
Storage
The product may be stored at 4°C for one month and at -20°C for long-term storage. After reconstitution, aliquot and store at -20°C.
Reconstitution
The powder should be reconstituted with 500 µl of reconstitution buffer. 5 µl is recommended for each lane of a mini-gel.
Application
The High Range EasyWestern protein standard is designed to help identify high-molecular-weight protein bands in Western blots when proteins are separated by low concentration gels or gradient gels. Because of the IgG binding ability, there is no need for additional reagents such as specific antibody or conjugate of streptavidin. It can be used as a convenient replacement of His-tagged markers, biotin-labeled markers, etc. It can bind total IgG from human, mouse, rat, sheep, goat, rabbit, etc. Because the marker binds to constant region of IgG, it is not compatible with Western blots where protein A or protein G conjugates are used as secondary detection reagents.
