Open Reading Frame (ORF) cloning provides easy and quick means to turn DNA sequences into expression-ready clones. GenScript ORF cloning service features advanced technology platform and GenPoolTM ORF clones database that currently hosts 1,124,477 ORF clones from 90 different species.
- GenPoolTM ORF clones database:
Our ORF clones database includes 1,124,477 ORF clones from 90 different species, and provides various convenient search options.
Research Areas-- 3,078 records
Protein Families-- 1,194 records
Signaling Pathways-- 2,087 records
Function Groups-- 3,782 records
- Gene-on-DemandTM gene synthesis technology platform:
Our proprietary technology platform is a combination of several advanced technologies, including GenPoolTM ORF clones database, OptimumGeneTM codon optimization software, and vector collection and clone archive database.
- Expression-ready cloning:
ORFs can be cloned into our pDream vector, that allows direct use in three different expression systems- bacteria, Sf9 cells, and mammalian cells without any
further subcloning. However, ORFs can be cloned into other commercial or custom vectors.
- DYKDDDDK-tagged cloning:
ORFs cloned into pDream vector are tagged with a DYKDDDDK tag, facilitating of its expression analysis with anti-DYKDDDDK antibody
(GenScript, A00013). This epitope tag is the most widely used
epitope tag employed in structural and functional studies of proteins.
The tag can be easily removed using enterokinase (EK, GenScript, Z01003).

- Sequence Confirmation:
The sequences of your gene will be confirmed base-by-base against the complete ORF sequence. We will screen GenPoolTM ORF clones collection of full-length ORF clones database if no appropriate plasmid template is available.
- De Novo Synthesis or Precise Amplification:
GenScript now offers Gene-on-DemandTM gene synthesis services for any genes that are currently not already in our ORF clones database.
Using gene synthesis technology platform, we can de novo synthesize any sequence of your interest or precisely amplified the target gene you submitted.
- Cloning into Vector:
The gene of your interest can be cloned into our pDream vector, or any commercial or non-commercial vector of your choice.
- QC by Sequencing:
Every clone will be fully sequenced before delivery to ensure 100% accuracy.
Delivery Specifications:
Our typical turnaround time is one week if the ORF clone in pDream vector meets your need. If the ORF clone of your interest is not physically available, or it needs to be cloned into a vector other than pDream, the turnaround time is about 14 business days for genes up to 1 kb. Our typical delivery package includes the following:
- 10 µg lyophilized destination vector containing the gene of your interest
- Stab culture of E. coli transformed with the destination vector containing the gene (only for US customers) of your interest
- Sequencing data of each clone (electronic version)
- Construct maps (electronic version)
- Quality assurance certificate
