Description: Taq Supermix is a ready-to-use mixture of DNA polymerase, salts, magnesium, and dNTPs, optimized for efficient amplification of templates in reactions. The user need only add template and primers, cutting the reaction setup time by half. The Taq Supermix (Cat. No. E00033) comes as three vials. Each vial contains about 1.5 ml of solution, sufficient for 100 reactions (45 µl each).
Taq Supermix contents: 22 mM Tris-HCl (pH 9.0), 11 mM KCl, 11 mM (NH4)2SO4, 2.2 mM MgSO4, 220 µM dNTP Mix, 0.11% Triton X-100, 50 U recombinant Taq DNA Polymerase/ml and stabilizer.
Protocol: Taq Supermix is optimized for a standard 50 µl PCR reaction, which uses 45 µl of 1.1 X Taq Supermix and 5 µl of template DNA and primer solution. The following protocol is suggested as a starting point and guideline for using 1.1 X Taq Supermix. We recommend assembling reactions on ice.
- Set up reaction tubes or plates on ice.
- Add the following components in any order.
- 45 µl of 1.1 X Taq Supermix
- Primers (200 nM final concentration of each primer is recommended
- Template DNA solution
- Mix components and cover with mineral oil if necessary.
- Cap reaction vessels and load in thermal cycler.
- Run cycling program.
