DescriptionBone Morphogenetic Protein (BMP)-2, human is a bone-growth regulatory factor and a member of the transforming growth factor-β (TGF-β) superfamily of proteins. BMP-2 is synthesized as large precursor molecules, which are cleaved by proteolytic enzymes. The active form can consist of a dimer of two identical proteins or as a heterodimer of two related bone morphogenetic proteins.
Recombinant human Bone Morphogenetic Protein-2 produced in E. coli is a homodimeric, non-glycosylated polypeptide chain containing 115 amino acids and having a molecular mass of 26,018 Da. rHuBMP-2 is purified by proprietary chromatographic techniques.

Specific ActivityrHuBMP-2 is fully biologically active when compared to standard. The ED50 as determined by the cytolysis of MC3T3-E1 cells is less than 50 ng/ml.
StorageLyophilized rHuBMP-2 although stable at room temperature for three weeks, should be stored desiccated at -18°C or below. Upon reconstitution, rHuBMP-2 may be stored at 4°C for short periods (two to seven days). For long-term storage, it is recommended that a carrier protein (0.1% HSA or BSA) be added. Avoid repeated freeze-thaw cycles.
FormulationrHuBMP-2 is lyophilized from a concentrated (1 mg/ml) sterile solution containing 10 mM sodium citrate pH 3.5.
ReconstitutionIt is recommended that the lyophilized rHuBMP-2 be reconstituted in sterile 20 mM HAc (acetic acid) not less than 100 μg/ml, which can then be further diluted to other aqueous solutions.
Sequence analysisThe sequence of the first five N-terminal amino acids has been found to be Met-Gln-Ala-Lys-His.
