| Full Name | | Bone Morphogenetic Protein (BMP)-7, human |
|
| Abbreviated Name-1 | BMP-7, human;rHuBMP-7; rHuBMP7 |
|---|
| Abbreviated Name-2 | Bone Morphogenetic Protein -7; Bone Morphogenetic Protein7 |
| Description | Cellular responses to Bone Morphogenetic Protein (BMP)-7, human are mediated by the formation of hetero-oligomeric complexes of type I and II serine/threonine kinase receptors. Human and mouse BMP-7 share 98% amino acid sequence identity. Recombinant human BMP-7 produced in E. coli is a monomeric, non-glycosylated polypeptide chain containing 139 amino acids and having a molecular mass of 15,679.97 Da. rHuBMP-7 is purified by proprietary chromatographic techniques. |
| Source | E. coli |
| M.W. | 15,679.97 Da |
| Purity | Greater than 95.0% as determined by: (a) RP-HPLC analysis (b) Anion-exchange FPLC (c) Analysis by reducing and non-reducing SDS-PAGE silver-stained gel analysis |
| Endotoxin Level | Less than 0.1 ng/µg (IEU/µg) of BMP-7 |
| Specific Activity | The ED50, calculated by the dose-dependant proliferation of murine BALB/c 3T3 cells (measured by 3H-thymidine uptake) is < 0.1 ng/ml corresponding to a specific activity of 1×107 U/mg. |
| Storage | Lyophilized Bone rHuBMP-7 remains stable at room temperature for 3 weeks, but it is best stored desiccated below -18°C. Upon reconstitution rHuBMP-7 may be stored at 4°C for up to seven days. For long term storage it is recommended that a carrier protein (0.1% HSA or BSA)be added. Avoid repeated freeze-thaw cycles. |
| Formulation | The protein is lyophilized from a concentrated (1 mg/ml) sterile solution containing 10 mM sodium citrate pH 3.5. |
| Reconstitution | The lyophilized rHuBMP-7 should be dissolved in sterile 18 MΩ-cm H2O with a final concentration not less than 100 µg/ml. It may be further diluted in other aqueous solutions. |
| Quantitation | Protein quantitation was carried out by two independent methods: 1. UV spectroscopy at 280 nm 2. RP-HPLC analysis using a standard solution of BMP-7 as a reference standard
|
| Applications | 1. Molecular standard (Western, ELISA) in studying secreted BMP-7. 2. Preparing antibodies for BMP-7 monomer. 3. Molecule standard in detecting secreted BMP-7 in reduced SDS-PAGE. |
| Sequence analysis | The sequence of the first five N-terminal amino acids has been found to be Ser-Thr-Gly-Ser-Lys. |
