DescriptionCellular responses to Bone Morphogenetic Protein (BMP)-7, human are mediated by the formation of hetero-oligomeric complexes of type I and II serine/threonine kinase receptors. Human and mouse BMP-7 share 98% amino acid sequence identity. Recombinant human BMP-7 produced in E. coli is a monomeric, non-glycosylated polypeptide chain containing 139 amino acids and having a molecular mass of 15,679.97 Da. rHuBMP-7 is purified by proprietary chromatographic techniques.

StorageLyophilized Bone rHuBMP-7 remains stable at room temperature for 3 weeks, but it is best stored desiccated below -18°C. Upon reconstitution rHuBMP-7 may be stored at 4°C for up to seven days. For long term storage it is recommended that a carrier protein (0.1% HSA or BSA)be added. Avoid repeated freeze-thaw cycles.
FormulationThe protein is lyophilized from a concentrated (1 mg/ml) sterile solution containing 10 mM sodium citrate pH 3.5.
ReconstitutionThe lyophilized rHuBMP-7 should be dissolved in sterile 18 MΩ-cm H2O with a final concentration not less than 100 µg/ml. It may be further diluted in other aqueous solutions.
QuantitationProtein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm
2. RP-HPLC analysis using a standard solution of BMP-7 as a reference standard
Applications1. Molecular standard (Western, ELISA) in studying secreted BMP-7.
2. Preparing antibodies for BMP-7 monomer.
3. Molecule standard in detecting secreted BMP-7 in reduced SDS-PAGE.
Sequence analysisThe sequence of the first five N-terminal amino acids has been found to be Ser-Thr-Gly-Ser-Lys.
