| Abbreviated Name-2 | B-type Natriuretic Peptide |
| Description | B-type Natriuretic Peptide (BNP), human produced in E. coli is a single, non-glycosylated, polypeptide chain containing 32 amino acids and having a molecular mass of 3,464 Da. rHuBNP is purified by proprietary chromatographic techniques. It is secreted by the ventricles of the heart in response to excessive stretching of myocytes (heart muscles cells) in the ventricles. BNP appears to be a useful marker of cardiovascular risk, even in people with no clinical evidence of cardiovascular disease. |
| Source | E. coli |
| M.W. | 3,464 Da |
| Purity | Greater than 95.0% as determined by the following methods: (a) RP-HPLC analysis (b) Anion-exchange FPLC (c) Reducing and non-reducing SDS-PAGE silver-stained gel analysis |
| Endotoxin Level | Less than 0.1 ng/µg (IEU/µg) of rHuBNP |
| Specific Activity | Activity is confirmed by reaction with monoclonal antibodies against different epitopes of NT-proBNP. |
| Storage | Although lyophilized rHuBNP can remain stable at room temperature for three weeks, it is best stored desiccated below -18°C. Upon reconstitution, rHuBNP may be stored at 4°C for up to one week and below -18°C for longer periods. For-long term storage, it is recommended that carrier protein (0.1% HSA or BSA) be added. Avoid freeze-thaw cycles. |
| Formulation | The protein is lyophilized from 0.4 ml PBS buffer containing 20 mM phosphate buffer and 0.6 mM sodium chloride. |
| Reconstitution | It is recommended that the lyophilized rHuBNP be reconstituted in sterile 18 MΩ-cm H2O of not less than 100 µg/ml, which can then be further diluted to other aqueous solutions. |
| Quantitation | Protein quantitation is carried out by two independent methods: 1. UV spectroscopy at 280 nm 2. RP-HPLC analysis using a calibrated solution BNP as a reference standard
