Specific ActivityThe ED50, calculated by the dose-dependant proliferation of monkey 4MBr-5 indicator cells has been found to be 20-40 ng/ml.
StorageAlthough lyophilized rHuHGF remains stable at room temperature for three weeks, it is best stored desiccated below -18°C. Upon reconstitution, rHuHGF may be stored at 4°C for up to one week and below -18°C for longer periods. For long-term storage it is recommended that a carrier protein (0.1% HSA or BSA)be added. Aviod freeze-thaw cycles.
FormulationThe protein is lyophilized from a concentrated solution containing 0.1 M L-Arginine and 10 mM Tris at pH8.
ReconstitutionIt is that the lyophilized rHuHGF be reconstituted in sterile 18 MΩ-cm H2O not less than 100 µg/ml, which can then be further diluted to other aqueous solutions.
QuantitationProtein quantitation is carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 1.83 as the extinction coefficient for a 0.1% (1 mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics)
2. RP-HPLC analysis, using a calibrated solution of HGF as a reference standard
