Specific ActivityThe product shows itself fully biologically active when compared to standard. Its ED50 as determined by the cytolysis of murine L929 cells in the presence of Actinomycin D is < 0.05 ng/ml, corresponding to a specific activity of 9×108 IU/mg.
StorageLyophilized rmTNF-alpha although stable at room temperature for three weeks, may be stored desiccated below -18°C. Upon reconstitution rmTNF-alpha should be stored at 4°C for up to seven days and for longer periods -18°C. It is also recommended that a carrier protein (0.1% HSA or BSA) be added for long-term storage. Avoid repeated freeze-thaw cycles.
FormulationThe protein is lyophilized from a concentrated (1 mg/ml) sterile solution containing 20 mM Tris-HCL, pH=8, 5 mM MgCl2 and 10% glycerol.
ReconstitutionIt is recommended that to reconstitute the lyophilized rmTNF-alpha be reconstituted in sterile 18 MΩ-cm H2O not less than 100 µg/ml, which can then be further diluted to other aqueous solutions.
QuantitationProtein quantitation was carried out by the following two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 1.24 as the extinction coefficient for a 0.1% (1 mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. RP-HP analysis
