| Full Name | | Pegylated Interferon-α 2b, human |
|
| Abbreviated Name-1 | PEG-IFN-α 2b, PEG-IFN a2b, PEG-IFNa2b, PEG IFNa2b, PEG IFN α2b, PEG IFN alpha 2b |
|---|
| Abbreviated Name-2 | PEG-interferon-α 2b, PEG-interferon-alpha 2b, PEG-interferon a2b, PEG interferon α2b |
| Description | At least 23 different variants of Interferon-α are known. The individual proteins have molecular masses between 19,000-26,000 Da and consist of proteins with lengths of 156-166 and 172 amino acids. All IFN-α subtypes possess a common conserved sequence region between amino acid positions 115 and 151, but the amino-terminal ends are variable. Many IFN-α subtypes differ in their sequences at only one or two positions. Naturally occurring variants also include proteins truncated by 10 amino acids at the carboxyl-terminal end. Recombinant Human Interferon-α 2b produced in E. coli is a single, non-glycosylated, polypeptide chain containing 165 amino acids and having a molecular mass of 19,269 Da. GenScript Pegylated Interferon-α 2b, human is manufactured by attaching a 40,000 Da NHS-mPEG to the primary amino acid of IFN-α 2b. |
| Source | E. coli |
| M.W. | 19,269 Da |
| Purity | Greater than 98.0% as determined by the following methods: 1. RP-HPLC analysis 2. Anion-exchange FPLC 3. Reducing and non-reducing SDS-PAGE Silver Stained gel analysis |
| Endotoxin Level | Less than 0.1 ng/μg (IEU/μg) of IFN-α 2b |
| Specific Activity | Pegylated IFN-α 2b is more biologically active than standard IFN-α. Its specific activity, as determined by viral resistance assay using bovine kidney MDBK cells, was found to be 5×107 IU/mg. |
| Storage | Although lyophilized PEG-IFN-α 2b remains stable at room temperature for three weeks, it is ideally stored desiccated below -18°C. Upon reconstitution, PEG-IFN-α 2b may be stored at 4°C for up to one week and below -18°C for longer periods. For long-term storage it is recommended that a carrier protein (0.1% HSA or BSA) be added. Avoid freeze-thaw cycles. |
| Formulation | Lyophilized from a (1 mg/ml) solution in containing 5.55 mg sodium phosphate dibasic, 5.55 mg sodium phosphate monobasic buffer, 296 mg sucrose and 0.37 mg Tween 80 |
| Reconstitution | It is recommended that the lyophilized PED-IFN-α 2b be reconstituted in sterile 18 MΩ-cm H2O not less than 100 μg/ml, which can then be further diluted to other aqueous solutions. |
| Sequence analysis | The sequence of the first five N-terminal amino acids has been found to be Met-Cys-Asp-Leu-Pro. |
