hook region A normally protected but reactive sequence of the 2-macroglobulin molecule, which is a scavenger for many proteinases in the blood. A proteinase can cleave a peptide bond in the protein's bait region, but then becomes susceptible to reaction with the thioester bond formed between a -carboxy group and a -thiol group. Once caught by this hook, the proteinase remains covalently bound to the protein, and is cleared with it from the circulation.
