S1 nuclease mapping

A technique for identification of nucleotides at the 5'-end of an mRNA which, because it retains at that end its original triphosphate moiety, may be specifically labelled for sequencing by the Maxam-Gilbert technique. A restriction fragment that is complementary to the 5'-end of the mRNA protects it against S1 nuclease digestion, which is specific for single-stranded RNA and DNA; the only remaining polynucleotides therefore are the 5'-end of the mRNA and its unlabelled complement.
