Description
GenScript Insulin-Like Growth Factor (IGF)-1, human is purified by proprietary chromatographic techniques. The insulin-like growth factors (IGFs) are polypeptides with high sequence similarity to insulin. IGFs are part of a complex system that cells use to communicate with their physiologic environment. Insulin-like growth factor 1 (IGF-1) is mainly secreted by the liver as a result of stimulation by growth hormone (hGH). IGF1 is important for both the regulation of normal physiology, as well as a number of pathological states, including cancer. Recombinant Human IGF-1produced in E. coli is a single, non-glycosylated, polypeptide chain containing 70 amino acids and having a molecular mass of 7,655 Da. rHuIGF-1is purified by proprietary chromatographic techniques.

Greater than 95.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Endotoxin Level
Less than 0.3 ng/µg (IEU/µg) determined by LAL test.
Specific Activity
The ED50, calculated by the dose-dependant proliferation of murine BALBC 3T3 cells (measured by 3H-thymidine uptake) is < 1.0 ng/ml, corresponding to a specific activity of 1×106 U/mg. For most in-vitro applications, IGF-I exerts its biological activity in the concentration range of 0.2-20 ng/ml.
Storage
Lyophilized rHuIGF-I although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution rHuIGF-I should be stored at 4°C between 2-7 days and for future use below -18°C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid repeated freeze-thaw cycles
Formulation
The protein was lyophilized after extensive dialysis against acetic acid.
Reconstitution
It is recommended to reconstitute the lyophilized rHuIGF-I in 100 mM Acetic acid not less than 100 µg/ml, which can then be further diluted to other aqueous solutions.
Quantitation
Sequence analysis
The sequence of the first five N-terminal amino acids was determined and was found to be Gly-Pro-Glu-Thr-Leu. N-terminal methionine has been completely removed enzymatically.
