Purified protein A is a highly stable cell surface receptor produced by several strains of Staphylococcus aureus. With a molecular weight of 35,000 Da, it is made up of a single polypeptide chain containing five Ig-binding regions. Purified protein A is ideal for purification of polyclonal or monoclonal IgG antibodies. Purified protein A binds to most human and mouse IgG subclasses (e.g. human IgG1, IgG2, IgG4; mouse IgG1, IgG2a, IgG2b, IgG3). It also binds to total IgG from cow, guinea pig, hamster, horse, pig, and rabbit. Protein A has little affinity to chicken IgY. To reconstitute the powder, for 10 mg size (Z02201-10), add 0.8ml water and 0.2ml 1xPBS, and the final concentration is 10mg/ml in 1x PBS. Please scale up accordingly for other sizes, such as Z02201-50 or Z02201-500.

Purified protein A may be conjugated with various reporter molecules, including fluorescent dyes (FITC), enzyme markers (peroxidase, β-galactosidase, alkaline phosphatase), biotin, and colloidal gold. These conjugates can be used to detect immunoglobulins in various immunochemical assays including Western blotting, immunohistochemistry, and ELISA applications. In addition, purified protein A may be immobilized on a solid support such as agarose or acrylic beads to purify polyclonal or monoclonal immunoglobulins. It can also be used in immunoprecipitation assays.
