Our siRNA Service Process:
  1. Use our Design Center to identify siRNA target sites and design the inserts.
  2. Send us the sequence of the designed DNA inserts.
  3. Select a GenScript siRNA vector or ship us any commercially available siRNA vector. For GenScript vectors, there is a one-time-only fee of $225 - $350 per vector.
  4. GenScript will synthesize the DNA insert, clone it into the specified vector, and verify it base-by-base by sequencing.
  5. The final plasmid (Miniprep or Maxiprep) will then be delivered with the sequence data and QC report.



Service Specifications:

Service Cat. No.: SC1007.
Description: A small DNA insert (about 70-80 bp) encoding a short hairpin RNA targeting the gene of interest is cloned into a siRNA expression vector. The insert-containing vector can be transfected into the cell, where it expresses the short hairpin RNA.
  • Promoter: Options include human U6 and H1 promoters.
  • Vector antibiotic resistance: Options include neomycin, hygromycin, and zeomycin.
  • Term: Poly (T) termination signal.
  • BamH I and Hind III are used to clone the DNA insert into GenScript pRNA vectors.
Quantity: 4 µg (MiniPrep) or 100 µg (MaxiPrep), and multi-milligram quantities are available.

Quality Control: The insert is sequence-verified, and trace data is provided together with the plasmid.
Protocol: Download PDF file (U6 or H1 promoter), PDF file (CMV promoter).
Storage: Store at -20 oC after delivery.

Delivery:

The typical delivery includes a construct map, approximately 4 μg of lyophilized plasmid DNA containing the siRNA construct, and sequence data documenting the DNA sequence of each clone. The typical delivery time is about three weeks.
