Firefly Luciferase siRNA
Constructs: siFLuc
siFLuc inserts are siRNA constructs designed to knock down
firefly luciferase. In GenScript's controlled experiments,
this siRNA can knock down firefly luciferase activity by about
80%. The target sequence (CTTACGCTGAGTACTTCGA) was selected
based on an article published in Nature Genetics (2002) 32:107. The target
sequence matches the pGL3-control and is designed to silence
firefly luciferase expressed by a co-transfected pGL3-control
vector (Promega, Cat. #E1741). The alignment is as follows:

siRNA constructs designed to knock down Renilla luciferase activity. In our controlled experiments, this siRNA can knock down Renilla luciferase
activity by about 80%. The target sequence
(GTAGCGCGGTGTATTATAC) was selected based on an article published in Nature Biotechnology (2002), 20:497. The
target sequence matches pRL-TK (Promega, Cat. #E2241) and is designed to silence Renilla
luciferase expressed by a co-transfected pRL-TK vector. The
alignment is as follows:

The
si-cGFP controls are siRNA constructs designed to knock down
cGFP (coral green fluorescent protein) expression. In our
controlled experiments, this siRNA
can knock down cGFP fluorescence by about 80%. The si-cGFP
controls are easy to use and allow scientists to monitor the
effects of siRNA by using fluorescence microscopes. The
si-cGFP inserts have been cloned into pRNA-U6.1/Neo and
pRNA-H1.1/Neo vectors. As such, they can be used to test the
effects of both U6 and H1 promoters.
