Tagged Protein Purification ResinsTo simplify the purification of recombinant proteins by affinity chromatography, a tag of protein/peptide or other molecule with known size is usually incorporated into the proteins. The two most commonly used tags are 6×Histidine and Glutathione S-transferase (GST). Biotin and Streptavidin are also frequently attached to proteins of interest to enable simple affinity purification. GenScript provides several affinity resins for convenient and reliable purification of tagged proteins. High Affinity Ni-Charged ResinThe most common tag is 6×His that strongly binds to divalent metal ions such as nickel and cobalt. GenScript High Affinity Ni-Charged Resin is 4% cross-linked agarose with metal-chelating nitrilotriacetic acid (NTA) groups covalently bound to its surface, and the resin is pre-charged with nickel and ready to use for batch/gravity purification of 6×His tagged proteins under native or denaturing conditions. The Low leakage of Ni2+ ions on the resin ensures reliable capture of 6×His tagged proteins in repeated purifications. Glutathione ResinGlutathione S-transferase (GST) is a roughly 26 kDa protein tag that is often incorporated into a protein of interest to facilitate purification of the GST fusion protein. GST strongly binds to its substrate, glutathione. GenScript Glutathione Resin is 4% cross-linked agarose with glutathione pre-coupled, designed for rapid and single-step purification of GST or GST fusion proteins under very mild condition. The high binding capacity and good flow property make the resin an excellent choice for batch or column gravity purification and scale-up. Streptavidin ResinGenScript Streptavidin Resin is prepared by covalently coupling highly purified streptavidin to 4% cross-linked agarose. This resin can be used for affinity purification of biotinylated proteins, antibodies, lectins, etc and as well as elimination of biotin and biotinylated molecules from samples. Anti-DYKDDDDK G1 Affinity ResinDYKDDDDK or FLAG® tag is an octapeptide tag that can be added to a protein using recombinant DNA technology. It can be fused to the N-terminus or C-terminus of a target protein to facilitate detection and purification. GenScript Anti-DYKDDDDK G1 Affinity Resin is designed for the purification of DYKDDDDK-tagged protein from commonly used protein expression systems including bacteria, yeast and mammalian cells. The DYKDDDDK-tagged protein in the cell lysate can specifically bind to the anti-DYKDDDDK monoclonal antibodies coupled to the resin.
