Adenoviral Vector Services
The adenovirus (AV) is a non-enveloped particle containing linear double-stranded DNA (dsDNA). AVs infect both dividing and non-dividing cells and replicate as episomal elements in the nucleus. There exist many AV serotypes. To date, the majority of adenovirus vectors are developed based on serotypes 2 and 5 by replacing either the E1 or E3 gene with a transgene, resulting in the loss of viral replicability. These recombinant viruses are replicable only in cells that express the E1 or E3 gene products at very high concentrations, creating a highly efficient, controllable viral vector system for therapeutic applications.
Service-Related Specifications (SC1166):
- Recombinant AV vector construction: GenScript's adenovirus vectors are standard E1- and E3-deleted AV vectors. Cloning plasmids are available to express the transgene as the single gene product or simultaneously with a GFP reporter under a bi-directional promoter for the purpose of transfection efficiency analysis. An inducible tetracycline adenovirus vector is also available for controlled gene expression.
- Recombinant AV production: Recombinant adenovirus vectors are derived from plaque-forming units (pfu). A single plaque is selected and amplified by serial infection of 293A cells on 20×15 cm plates after transduction with the linearized vector plasmids. The recombinant AVs are purified, titered, and verified by restriction digestion and sequencing.
- Recombinant AV characteristics: Routinely, a single adenovirus vector virus stock is prepared starting from 30×150 mm-cell culture dishes.
- Yield: 1×103 viral particles/cell
- Biological titer: 108-1010 pfu/ml(concentrated)
- Final volume: 1 ml of purified viruses in phosphate buffered saline (PBS)
- Storage: The AV vector batches are best stored at -80°C
