Hsp104 is a protein disaggregase from yeast that we have engineered to counter the misfolding of proteins that aggregate in ALS and Parkinson's disease.
Numerous missense mutations, both conservative and non-conservative, can potentiate Hsp104.
By screening large libraries of Hsp104 variants that comprehensively cover sequence space we aim to uncover variants with improved properties, and also learn more about the mechanism of Hsp104 potentiation.
