For the purification of GPR15LC11-GPR15-Gi-scFv16 complex, GPR15LC11 (WVVPGALPQV) was synthesized by GenScript using solid-phase peptide synthesis, the purity of the synthesized peptide was determined to be 95%, cell pellets from 1L culture were thawed at room temperature and resuspended in low salt buffer containing 20 mM HEPES pH 7.4, 100 mM NaCl, 5 mM CaCl2, 5 mM MgCl2, 10% glycerol, protease inhibitor cocktail (Thermo Fisher). |
