Description Nicastrin is a Type I transmembrane glycoprotein. Along with presenilin, APH-1, PEN-2, it comprises the multimeric gamma-secretase complex. The gamma-secretase complex can cleave the beta-amyloid (A4) precursor protein and yields amyloid beta peptide, the main component of the neuritic plaque and the hallmark lesion in the brains of patients with Alzheimer's disease. Nicastrin is also thought to be involved in cell proliferation and signaling, especially in regards to activation of Notch receptors as loss of nicastrin expression results in mouse embryonic lethality.
GenScript Rabbit Anti-Nicastrin Polyclonal Antibody is developed in rabbit hosts using a synthetic peptide, FTRDLMEKLKGRTSRIAGLAVSL, corresponding to amino acids 103-125 of the N-terminus of human nicastrin.
Rabbit Anti-Nicastrin is highly purified from rabbit antiserum by immunoaffinity chromatography and is supplied as a 40 μg aliquot at a concentration of 0.5 mg/ml.

Specificity GenScript Rabbit Anti-Nicastrin Polyclonal Antibody can detect endogenous level of human nicastrin. Predicted to react with mouse and rat nicastrin according to sequence homology.
Storage The antibody is stable for 2-3 weeks if stored at 2-8°C. For long term storage, aliquot and store at -20°C or below. Avoid repeated freezing and thawing cycles.
Note Small volume of antibody will occasionally become entrapped in the cap of the vial during shipment and storage. Before use, centrifuge the vial briefly to bring down any liquid in the cap.

Working concentrations for specific applications should be determined by the investigator. Appropriate concentrations will be affected by several factors, including secondary antibody affinity, antigen concentration, sensitivity of detection method, temperature, and length of incubations, etc. The suitability of this antibody for applications other than those listed below has not been determined. The following concentration ranges are recommended starting points for this product.
ELISA: 0.05-0.2 µg/ml
Western blot: 0.1-1.0 µg/ml
Western Blot Using ONE-HOUR WesternTM Kit: For quick results, we recommend the GenScript ONE-HOUR WesternTM Kit L00204C. Mix 15 µg of GenScript Rabbit Anti-Nicastrin Polyclonal Antibody with 10 ml of WB solution for 8 cm X 8 cm membrane.
