Bacterial Expression System

Bacterial Protein Expression System is the most common and economical mean of protein expression available today. However, two major problems often prevent researchers from accessing the system's full advantages. One problem is that the protein of interest cannot always be expressed using a specific vector/strain combination. A second problem is the occasional insolubility of the proteins expressed in a bacterial system.

Over the years, GenScript has developed BacPower™ and FoldArt™ technologies to tackle hard-to-express and hard-to-dissolve proteins. With BacPower™ technology, the expression level can be greatly improved for most proteins. In addition, our FoldArt™ protein refolding technology is designed to determine the optimal refolding conditions by evaluation in a series of buffers. 1. Protein refolding

Overproduction of proteins in E.coli can cause the formation of inactive, misfolded and insoluble protein aggregates. GenScript provides proprietary refolding technology to solve this problem. A case study shows that over 95% of inclusion bodies are solublized and refolded. The purity of the refolded protein is more than 85% and is delivered in customized buffer.

2. Tag removal

Presence of epitope tag in recombinant proteins may result in changes in protein structure, toxicity, and loss of bioactivity. To avoid these problems, tag removal after protein expression is requested by some customers as an additional service. Shown below is protein that is cleaved by SUMO protease and successfully isolated. Recovered target protein is more than 90% pure and 80% tag free.

Protein sequences are subject to pre-project analysis. Exclusions are applicable for membrane proteins and toxic proteins.

For eligible proteins, a minimum of 3mg soluble protein with 75% or 85% purity will be delivered to customers. The protein services will be free of charge if GenScript's evaluation reveals that the expression level is insufficient to eventually obtain 3mg of purified protein. To increase the expression level of your protein, gene optimization using our OptimumGene^™ technology is recommended. Up to 3 fusion tag(s) may be tested to increase the expression level and solubility of the protein and are applied for protein production and purification. Fusion tags may be removed by introducing an optional protease cleavage site between the tag and the target protein (note: tag removal service is offered at extra cost) . Insoluble proteins will be subject to refolding using our FoldArt^™ technology at no additional fee.
