This peptide mimicking the NS5A-5B cleavage junction of the nonstructural protein (NS) is an efficient substrate used by Shimuzu et al. to detect NS3 protease activity in a standard assay (Km = 411 mM and Kcat = 0.14 min-1, without NS4 activator). Cleavage occurs between Cys and Ser. The cleavage products are analyzed by RP-HPLC.
