Description |
Human Interferon alpha 2a (IFN alpha 2a) produced in E. coli is a single, non-glycosylated, polypeptide chain containing 165 amino acids and having a molecular mass of 19,241 Da. The Interferon alpha 2a (IFN alpha 2a) gene was obtained from human leukocytes. The Recombinant Human Interferon alpha 2a (IFN alpha 2a) is purified by proprietary chromatographic techniques. |

Storage |
Lyophilized Recombinant Human Interferon alpha 2a (IFN alpha 2a) remains stable at room temperature for
three weeks, but it is best stored desiccated below -18°C.
Upon reconstitution recombinant Human Interferon alpha 2a (IFN alpha 2a) should be stored at 4°C for up to seven days. For long term storage it is recommended that a carrier protein (0.1% HSA or BSA ) be added. Avoid repeated freeze-thaw cycles. |

Lyophilized from (1 mg/ml) solution containing 7.21 sodium chloride and 0.77 mg ammonium acetate. |
Reconstitution |
It is recommended to reconstitute the lyophilized rHuIFN-α 2a in sterile 18 MΩ-cm H2O not less than 100 µg/ml, which can then be further diluted to other aqueous solutions. |
Quantitation |
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.924 as the extinction coefficient for a 0.1% (1 mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a calibrated solution of IFN-α 2a as a Reference Standard.

Sequence analysis |
The sequence of the first five N-terminal amino acids was determined and was found to be Cys-Asp-Leu-Pro-Gln, conforming to the sequence of native human IFN-α. N-terminal methionine has been completely removed enzymatically.
