One-Step WesternTM Kit Accessories

GenScript is pleased to provide reagents and other accompanying products for its line of One-Step WesternTM Kits, including detection substrates, solutions, membranes, PAGE gels, and labware.

The detection substrates include Chemiluminescent HRP Substrate and ChromoSensor TMB Substrate. The solutions are wash buffer, pretreat solutions, A&G blocker, and stripping solutions. The PAGE gel is our ExpressTM Precast Gel. The labware are forceps, dot boxes, and wash boxes.


Example:

1. Shown below is a western blot detection of housekeeping gene product GAPDH protein using LumiSensorTM Chemiluminescent HRP Substrate Kit.
Western blot using the HRP substrate Figure 1. 10 μg, 5.0 μg, 2.5 μg, 1.25 μg, 0.62 μg, 0.31 μg, 0.16 μg, and 0.08 μg of Hela cell lysate (BD Biosciences, 611449) were loaded into Lane 1, Lane 2, Lane 3, Lane 4, Lane 5, Lane 6, Lane 7, and Lane 8, respectively. The blot was developed with the LumiSensorTM Chemiluminescent HRP Substrate Kit.

3. Comparison of ExpressTM Precast Gels with other Precast Gels for Western Blot detection of His-tagged proteins.
precast gels Figure 2. 10 μl, 5.0 μl, 2.5 μl, 1.3 μl，and 0.6 μl of 6xHis Protein Ladder (Qiagen, Cat. No. 34705, reconstituted in 250 μl 1X SDS-PAGE sample buffer as suggested by Qiagen) were loaded in Lane 1, Lane 2, Lane 3, Lane 4, and Lane 5 of a 4 - 20% ExpressTM PAGE Gel, respectively. After electrophoresis, the His-tagged proteins were transferred to nitrocellulose membrane and then detected using One-StepTM His-Tag Western Detection Kit (L00212). This experiment was repeated by using a 4 - 20% precast gel (Brand A) from company A.
