Description
The secretin receptor SCTR is Gs-coupled GPCRs expressed in the duodenum, jejunum, pancreas, tertiary bronchus, lung parenchyma and pulmonary artery. There appears to be important species differences in the cellular and organ distribution of secretin and secretin family receptors. The best example of this is the confusing series of observations in pancreas of guinea-pig, rat, and human. There are also differences in the pharmacologic profile of secretin receptors from different species, despite minimal changes in primary structure.

Figure 1. Human Secretin-induced concentration-dependent stimulation of intracellular calcium mobilization in CHO-K1/SCTR/Gα15 and CHO-K1/Gα15 cells. The cells were loaded with Calcium-4 prior to stimulation with an SCTR receptor agonist, Human Secretin. The intracellular calcium change was measured by FlexStation. The relative fluorescent units (RFU) were plotted against the log of the cumulative doses (10-fold dilution) of Human Secretin (Mean ± SD, n = 2). The EC50 of Human Secretin on SCTR co-expressing with Gα15 in CHO-K1 cells was 9.6 nM. The S/B of Human Secretin on SCTR co-expressing with Gα15 in CHO-K1 cells was 17.
