Description
The melanocortin receptor 3 (MC3 receptor) is Gs-coupled GPCRs expressed in brain, placental, and gut tissues but not in melanoma cells or in the adrenal gland. MC3 receptor knockouts exhibit a metabolic syndrome. At 4-6 months old, MC3 receptor knockout mice show increased fat mass, reduced lean mass and a higher feed efficiency, with normal metabolic rates. MC3 knockout mice are hyperleptinaemic and males are often mildly hyperinsulinaemic. GenScript's cloned human MC3R-expressing cell line is made in the 293/CRE-Luciferase host.

Figure 1. Drug-induced concentration-dependent stimulation of intracellular cAMP in 293/MC3/CRE-Luc cells. After stimulation with MC3 receptor agonist, ACTH and NDP-α-MSH, the cells are checked with Bright-GloTM Luciferase Assay System and the relative luminescence units (RLU) of luciferase were recorded on a FlexStation. The RLU were plotted against the log of the cumulative doses (10-fold dilution) of ACTH and NDP-α-MSH (Mean ± SD, n = 2). The EC50 of ACTH and NDP-α-MSH on MC3 co-expressing with CRE-Luc in 293 cells was 0.18 nM and 8.7 nM, respectively. The S/B of ACTH and NDP-α-MSH on MC3 co-expressing with CRE-Luc in 293 cells was 21.
