    This kit has been validated for the suitability of AAV serotypes 1, 2, 5, 6, 8, and 9, and the typical working range can be found in the table below:

    Note: The materials chosen for the validation of the working range for each serotype in the table include AAV2 virus-like particles (VLPs) produced by GenScript and empty capsids from Progen for the remaining serotypes.

    The antibody in the kit could be recognize AAV2.7m8, AAVDJ and AAVrh.1, but the kit is not validated for the detection of the three AAVs. Users can follow the protocol of this kit to evaluate the suitability of other serotypes.

  • Does the AAVX Titer Capsid ELISA Kit - Multiple Serotypes provide a positive standard?

    This kit only provides a standard for AAV2, a virus-like particle (VLP) produced by GenScript. Users need to establish other AAV serotypes standards for standard curves and quality control.

  • If users do not have positive standards for other serotypes of AAV, how to use the AAVX Titer Capsid ELISA Kit - Multiple Serotypes to quantify the titer of other AAV serotypes?

    It is recommended to use a positive control that matches the AAV capsids in the samples to generate an accurate standard curve for quantification. Please note that this kit provides standards for AAV2 only; standards for other serotypes are not included.

    If users are unable to obtain positive standards for other AAV serotypes, we provide a conversion method that using the AAV2 standard in the kit to establish a standard curve and determine the concentration of the samples. The determined concentration can be used as X in the following formula to calculate the corresponding titer for the serotypes. The formulas in the table have been validated through correlation. Please note that this method is intended for rough estimation in research use only.

  • How many samples can be tested with one ELISA kit?

    This kit has a pre-coated 96-well microplate, which contains 8 wells x 12 strips. We recommend diluting the standard curve gradient according to our manual and preparing 8 working solutions for the standard. For accurate quantification, it's suggested to test both standards and samples in duplicate. This procedure will yield 40 replicate measurements once the standard curve is applied.

  • Can additional washing reagents be provided?

    Users could purchase the Wash Solution (GenScript, B00063) that is compatible with this assay kit separately.

  • Is it effective to measure values beyond the assay working range?

    No, it is ineffective. The dilution factor needs to be adjusted and the measurement should be repeated.
