Pelham hypothesis

The suggestion that aggregation of a globular protein occurs when the protein is unfolded, for instance by heat, and the specific intramolecular interactions that determined its native structure are replaced by the same interactions with other unfolded molecules of the same protein. In this view, heat-shock proteins help prevent this aggregation by competing with these intermolecular reactions and by somewhat weaker interactions, maintain the partially unfolded protein in an unaggregated state.
