• 2’Ome (the first 3 bases) at 5’ end of crRNA was found compromising Cpf1 activity (McMahon et al., 2018; Rozners, 2022; Ha et al, 2020)
• 2’Ome (the last 3 bases) at 3’ end was well tolerated, showing no negative impact on editing activity (McMahon et al., 2018; Ha et al, 2020). A report demonstrates the modification suppressed Cas12a‘s affinity to off-target DNA while maintaining high affinity to on-target DNA (Ke et al., 2022).
