Product Description

The cAMP Detection Kit (Cell-Based Assay) is a competitive immunoassay designed for the sensitive and accurate quantification of intracellular cyclic adenosine monophosphate (cAMP) levels. This assay enables real-time monitoring of dynamic changes in intracellular cAMP concentrations.
In the assay system, cAMP-A2 (acceptor) specifically binds to Anti-cAMP-Eu (donor), forming a donor–acceptor pair capable of energy transfer. Upon excitation at 320 nm, the europium-labeled donor emits fluorescence at 620 nm, which in turn excites the acceptor, resulting in secondary emission at 665 nm. The ratio of fluorescence intensities at 665 nm to 620 nm serves as an indicator of the extent of donor–acceptor binding.
Endogenously produced cAMP or unlabeled cAMP competes with cAMP-A2 for binding to Anti-cAMP-Eu. As a result, an increase in intracellular cAMP levels leads to reduced donor–acceptor complex formation, thereby decreasing the 665 nm/620 nm fluorescence signal ratio. Thus, the intensity of the fluorescence signal is inversely proportional to the concentration of intracellular cAMP.

Storage

The kit should be stored and transported at –20°C.
● After first use, cAMP Standard, Anti-cAMP-Eu, and cAMP-A2 should be aliquoted as needed and stored at –20°C to avoid repeated freeze-thaw cycles.
● Stimulation Buffer and Lysis Detection Buffer should be stored at 4°C.
