| Product Description |
VHH, also known as nanobodies, are the smallest
functional fragments of naturally occurring heavy-chain antibodies found in
camelids. Their compact size, high stability, and solubility make them valuable
tools for various applications in biotechnology, diagnostics, and therapeutics.
Besides, when VHH are intended for use in humans, it is often necessary to
humanize them to reduce their immunogenicity and enhance their compatibility
with the human immune system. Therefore, the purification of both humanized and
camelid VHH are crucial. GenScript’s MonoRab™ Anti-VHH Affinity Magnetic
Beads (Cat. No. L01034) is specifically designed for purifying camelid
and humanized VHH from commonly used protein expression systems such as
bacteria, yeast, and mammalian cells. The VHH in cell lysate can specifically
bind to the anti-VHH antibody coupled to the beads. Non-specific binding
reactions are eliminated through rigorous washing steps, and the purified target
protein is eluted with a high recovery rate. |

Regeneration performance of MonoRab™ Anti-VHH Affinity Magnetic Beads
The target VHH was captured by GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads (Cat. No. L01034). The 100 μL settled beads were washed twice with 1 mL TBS and then eluted with 1 mL acidic eluent (0.1 M glycine HCl, pH 2.5). Subsequently, the beads were regenerated by 1 mL regeneration buffer for reuse. The coefficient of variation (CV) of the amount of eluted target protein was less than 20% for a total of 10 replicate purifications indicating that GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads is capable of multiple repeated use with relatively high consistency when eluted by acidic eluent. »
Purification of humanized VHH from E.coli whole cell lysate
The humanized VHH (M.W.=15 kDa) from E.coli whole cell lysate was captured by GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads (Cat. No. L01034). The captured humanized VHH was subsequently eluted using an acidic eluent (0.1 M glycine HCl, pH 2.5) and subjected to SDS-PAGE analysis. The results showed that GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads are suitable for high-specificity purification of humanized VHH from E.coli whole cell lysate

Purification of camelid VHH from various lysates
The camelid VHH (M.W.=15 kDa ) from E.coli whole cell lysate, yeast whole cell lysate or mammalian cell culture supernatant was captured by GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads (Cat. No. L01034). The captured camelid VHH was subsequently eluted using an acidic eluent (0.1 M glycine HCl, pH 2.5) and subjected to SDS-PAGE analysis. The results showed that GenScript’s MonoRab™ Anti-VHH Affinity Magnetic Beads are suitable for high-specificity purification of camelid VHH from widely used protein expression systems.
