Principle

1. What’s Strep-tag^®？

The Strep-tag^® (WSHPQFEK) and Twin-strep-tag^® (WSHPQFEKGGGSGGGSGGSAWSHPQFEK) are widely adopted affinity tags for the purification of recombinant proteins in basic research, diagnostics, and biomanufacturing. The Strep-tag^® is well suited for the routine purification of proteins expressed at moderate levels, whereas the Twin-strep-tag^® provides significantly enhanced binding affinity. This makes it particularly advantageous for isolating low-abundance targets, membrane-associated proteins, or labile protein complexes under challenging conditions.

Purifying His-tagged proteins is challenging, often requiring 2-3 complex steps to achieve desired purity, posing difficulties in process development and scale-up. In contrast, StrepCaptureXP’s ultra-high specificity enables one-step affinity chromatography to achieve high purity, significantly reducing time, labor, and costs, making it a more efficient purification method.

Enzymes involved in nucleic acid and metabolism require Mg²⁺ for activity, but His-tags can disrupt this by chelating metal ions, reducing enzyme function. StrepCaptureXP is a better choice for purifying enzymes while preserving activity for research.

3、Strep-tag^® vs His tag Application

Based on high-throughput experimental methods, three parallel tests were conducted on the purity performance of Strep-tagged proteins (purified using StrepCaptureXP Magnetic Beads) and His-tagged proteins (purified using Ni Magnetic Beads). The results are as follows:
