Yes. The TurboCHO™ transfection workflow is largely similar to Competitor T CHO Kit workflows, and we provide a step-by-step comparison table for reference. The key difference is that TurboCHO™ recommends a spin-down step prior to transfection, which enables higher cell density and improved titers.
We do not recommend the low-density dilution method, as it typically results in reduced productivity. As an alternative for automation-friendly workflows, customers can use a wave reactor to achieve high cell density and then dilute to the desired level prior to transfection while maintaining performance. Detailed protocols can be provided upon request.
The recommended incubation time is 1–1.5 minutes. It can be extended up to 8 minutes, but yields may decrease by approximately 10–15%.
A temperature shift is required after Day 1 feeding.
TurboCHO™ performs best when cells are centrifuged and resuspended to a high density before transfection. This step improves transfection efficiency and increases titer.
The same expression medium is used for both maintenance and transfection.
A temperature shift is required after Day 1 feeding.
An optional second feed may be performed on Day 5. Current recommendations are based on maintaining consistency with our validated 5-day and 7-day expression workflows.
The recommended DNA amount is 2 μg/mL, as specified in the protocol. Potentially, 1 μg/mL target DNA supplemented with 1.5 μg/mL filler DNA may achieve comparable results.
For large-volume processing, we recommend using Nalgene™ 2 L Bio Bottles for batch centrifugation. A large-capacity centrifuge can be used to process multiple bottles simultaneously.
