A BRAVE Approach for Generating Novel AAV Vectors


Engineering Adeno-associated viral (AAV) vectors through directed evolution has generated a plethora of potent novel capsids with improved tropism and function. However, the selection requires multiple generations of screening until the real functional capsids surface. Due to the truly random nature of the process, it is also inherently unreproducible, and the resulting capsid variants provide little mechanistic insights into the molecular targets engaged. In this webinar, we will investigate a novel method for rational capsid evolution called Barcoded Rational AAV Vector Evolution (BRAVE), which enables the selection of novel capsid structures using a single-generation screen.

In this webinar, we will cover:

  • Development of a novel high-throughput approach to engineer AAV capsids using only a single screening step
  • How oligo pools can be used in rational design libraries
  • How BRAVE screening can be used in vivo to identify novel motifs with improved viral function and elucidate neural function

Presenter’s Biography:

Marcus Davidsson received his Ph.D. in Neurobiology from Lund University in Sweden. During his doctoral studies, he worked in the laboratory of Dr. Thomas Bjorklund on developing methods for DNA barcoding and how to apply this technique to generate AAV vectors with tailored functions in order to study biological functions in the brain. He is currently a postdoctoral fellow in the lab of Dr. Fredric Manfredsson at Michigan State University. His current research interests focus on DNA barcoding, gene therapy, and AAV capsids.
