Job Title: Next-Gen BsAb Manufacturing – How TurboCHOTM Ensures High Productivity & Streamlined Purification

Abstract:

Recent advances in transient gene expression and AI-assisted antibody engineering have enabled rapid generation of a large number of bsAb formats with diverse structural architectures. However, the expression and purification of bsAbs remain technically challenging due to issues such as imbalanced chain pairing, misassembled species, and purification complexity. To address these bottlenecks, we have developed TurboCHOTM platform, a high-performance transient expression and purification platform tailored for next-generation bsAb production.

The platform is built upon an engineered CHO host with optimized process conditions, enabling robust and high-titer expression across a wide range of bsAb formats. To ensure correct chain pairing, transfection conditions are fine-tuned through rational plasmid ratio design. In the downstream process, we apply format-specific affinity resin selection to efficiently separate correctly assembled bsAbs from side products. Further, LC-MS analysis is integrated into the workflow for precise identification of mispaired species, providing critical data for purification. Final polishing steps, including SEC, IEX, HIC, or mixed-mode chromatography, are selected based on molecule-specific characteristics to achieve high purity suitable for downstream studies.

The TurboCHOTM platform streamlines bsAb production from expression to analytical release, supporting rapid molecule evaluation with consistent quality and scalability.

Since 2017, Dr. Segaliny has led innovative assay biology initiatives at Amberstone Biosciences, advancing a cutting-edge microfluidic-based single-cell screening platform for immunotherapeutics, with a focus on bispecific antibodies.
