GenScript Target Enrichment Solutions
As whole genome NGS always leads to high cost, long experiment time and large data storage space, scientists develop target enrichment technology to overcome these shortages when they only want to focus on the specific regions of interest. This method not only largely reduces cost but also makes the data analysis much easier for researchers.
Currently, two main target enrichment platforms are used in NGS, one is hybridization-based enrichment and the other is amplicon-based enrichment. Performance of GenScript’s double-stranded capture probes versus commercially available single-stranded probes
Sequences from a predesigned commercial panel of ~8,000 probes against 127 genes were synthesized by GenScript using an array oligo synthesis platform to create 120 mer, double-stranded DNA (dsDNA) capture probes. For comparison, individually synthesized, single-stranded DNA (ssDNA) probes with the same sequences were obtained from a competing vendor. A DNA library created from a commercial available standard sample was enriched for a 800 kb target region using either capture panel of DNA probes.
Deep, uniform sequence coverage of targeted regions.
Side-by-side comparison of GenScript’s dsDNA capture probes to column-synthesized ssDNA probes (Vendor T) revealed no difference in sequencing coverage depth, indicating comparable quality. An additional panel of dsDNA probes with the same sequences showed similar performance, indicating consistency between synthesized panels.
High reproducibility of capture efficiency.
Comparison of sequencing results between two technical replicates reveals low variation and high reproducibility using GenScript’s dsDNA probes.
