In biology, anastomose means to join or form a connection between structures. For example, in plants, the connections formed between leaf veins or branches and the connections between arteries, veins, or even between organs, such as the gastrointestinal tract in humans and animals, represent anastomoses. Anastomose may also refer to surgical procedures that join organs or vessels, exemplified by intestinal and vascular anastomosis. During embryonic development, anastomosis is one of the stages involved in new blood vessel formation (i.e., sprouting angiogenesis). Although the mechanisms involved in vessel anastomosis are not entirely elucidated, several molecular factors and pathways have been identified to play critical roles, including Flt1 (VEGFR1), VEGFA signaling, Wnt/β-catenin signaling, VE-cadherin, and Notch signaling. Biomaterials, such as hydrogels scaffolds that promote integrin binding, specifically α3/α5β1, induce endothelial sprouting, branching, and eventual vessel anastomosis supporting their potential application in vascular repair (e.g., post-stroke). Formation and remodeling of vasculature, including the process of anastomosis, may be replicated using organ-on-chip technology.
