Asparagine is an uncharged polar amino acid. The asparagine-hydroxylation modulates interaction with other activators, whereas the proline-hydroxylation targets HIF-1 for destruction in proteasomes. Asparagine deamidation is another PTM that must be monitored for changes over time. Deamidation entails the chemical cleavage of the amide group from the asparagine side chain, typically converting that residue to an aspartic acid or isoaspartic acid. As with isomerization, this degradation can be more impactful if the modification occurs on a CDR residue (or the reactive site of a recombinant protein) because it risks affecting activity. Asparagine is a nonessential amino acid used in the biosynthesis of proteins. The precursor of asparagine is OAA. Deamidation of asparagine and glutamine can also occur during sample preparation, and deamidation is often listed as frequent posttranslational modification in error-tolerant searches. Moreover, asparagine and lysine share a twofold degenerate codon box in the Universal Genetic Code.
