Overview

Cyclic peptides are emerging as a promising class of therapeutics, combining the desirable features of small molecule drugs and large biological molecules, such as high stability, potency, and selectivity. Compared with straight-chain peptides, cyclic peptides have better conformational stability, target affinity, and improved selectivity. Their ring-shaped structure, created through covalent bonding within the peptide chain, enhances their ability to target challenging protein-protein interactions implicated in various diseases, including cancer, infectious diseases, metabolic and neurodegenerative disorders, and autoimmune conditions. Despite synthesis and bioavailability challenges, advances in chemical ligation, molecular engineering, and peptidomimetics are paving the way for their development. Our platform offers a full spectrum of services for cyclic peptide drug development, from high-throughput screening and optimization to cGMP-compliant manufacturing, to streamline the transformation of these compounds from laboratory concepts to clinical applications.

Types of Cyclic Peptides

The -NH2 and -COOH groups on a peptide can form lactam through conjugation, resulting in a more stable cyclic conformation. Cyclosporine, Anidulafungin are these peptides. GenScript can provide various kinds of lactam peptides.

Disulfide cyclization is the most common peptide modification. FDA has approved many disulfide peptide drugs, such as Octreotide, Lanreotide and Linaclotide. GenScript can provide various complex disulfide cyclic peptides.

Click chemistry could link two units together through the formation of 1,2,3-triazole scaffold. This functional group has good biocompatibility. GenScript can provide conventional and copper-free click chemistry.

The conformational constraint of bicyclic peptides confers attractive drug-like properties, including the potential for high affinity to the target. GenScript has extensive experience in synthesizing bicyclic peptides with various center scaffolds.

Compared to the peptides with natural α helix, the α helix conformation of staple peptides is more stable and cell permeable. GenScript can provide various staple peptides, such as i,i+4 and i,i+7 staple peptides that are formed by RCM reaction.

Lactone peptides are abundant in nature and are an important source of drug discovery. Daptomycin, which has good efficiency on infections caused by Gram-negative organisms, is a natural lactone peptide. Genscript can provide various customized lactone peptides.
