sgRNA stands for "single guide RNA" or “small guide RNA”. It is a type of RNA molecule used in the CRISPR-Cas9 gene editing system, which is a powerful tool for making precise changes to DNA sequences in living cells. In the CRISPR-Cas9 system, the sgRNA molecule is designed to specifically bind to a target DNA sequence and guide the Cas9 protein to that location. Once the Cas9 protein is attached to the DNA, it can cut the DNA at the desired location, which allows for the addition, removal, or replacement of specific DNA sequences. The design of the sgRNA molecule is a critical step in the CRISPR-Cas9 gene editing process. It must be engineered to specifically recognize and bind to the target DNA sequence while avoiding unintended off-target effects. The sgRNA typically consists of a short sequence of RNA called the guide sequence, which is complementary to the target DNA sequence, as well as other RNA sequences necessary for proper function within the CRISPR-Cas9 system.
