sequencing by hybridization

Sequencing by hybridization is a proposed approach to rapidly sequencing genomic DNA. The critical tool for analysis is a superchip, a surface over which are attached unlabelled oligonucleotide probes.

Sequencing by hybridization (SBH) is an approach whereby overlapping oligonucleotide sequences are assembled to determine an organism's DNA sequence. Through the efficient method of sequencing by hybridization, scientists can gather information on the genomes of different species and organisms for the future development of biological sciences, medicine, and agriculture. Sequencing by hybridization is based on the complementary DNA strands’ ability to renature after melting, as reported by Doty.

Sequencing by hybridization is similar to CGH but is designed to detect mutations, taking advantage of the fact that nucleic acid hybridization conditions can be developed so that the intensity of hybridization of a target nucleic acid to a short oligonucleotide is significantly higher when the probe sequence is perfectly complementary to the target than when a single-base mismatch exists.
